| 簡介 | 該細胞系是從一名急性髓系白血病(AML)患者的外周血中建立的。這是一個8號21號染色體易位的白血病細胞系。這種 易位將AML1與ETO(或MTG8)基因并列 ，產生融合基因AML1-ETO(也稱為AML1-mtg或RUNX1-CBF2T1)。因 此 ，細胞產生嵌合的AML1-ETO蛋白。該蛋白下調CEBPA mRNA、蛋白和DNA結合活性 ，這對粒細胞的分化至關重 要。在增殖實驗中 ，培養的細胞對白細胞介素-3 (IL-3)、白細胞介素-6、粒細胞集落刺激因子(G-CSF)、粒細胞頂體噬 菌體CSF (GM-CSF)有反應 ，但對IL-1和IL-5無反應。該細胞復蘇后需要兩周左右恢復正常生長。 |

| 備注 | 該細胞為懸浮細胞 ，請注意離心收集細胞懸液 ，請勿直接倒掉細胞培養液 ，a) 該細胞復蘇后成活率較低 ，會出現大量 死細胞和死細胞碎片 ，培養兩周后有所好轉。建議每1-2周對細胞進行1000rpm ， 5mins離心 ，棄掉上清 ，加入新鮮 培養液 ，可以去掉部分細胞碎片和顆粒。培養過程中會出現死細胞和細胞碎片 ，收到郵寄的活細胞的用戶若發現 培養物內有部分死細胞和細胞碎片 ，此為正常現象。b) 細胞培養過程中會有輕微聚團 ，輕輕吹打開即可。當細胞密度 較大或者培養液變黃時 ，需要及時進行半換液或者換液。c) 該細胞對血清質量較為敏感 ，我庫建議您使用進口大 品牌優質血清進行培養。d) 請注意保持細胞密度在合適的范圍（3x105 ~ 3x106 /ml） ，不能過稀。 |
