美國Bio-rad伯樂脈沖場電泳儀
美國Bio-rad伯樂脈沖場電泳儀(PFGE，Pulsed Field Gel Electrophoresis)是一種分離大分子DNA的方法。在普通的凝膠電泳中，大的DNA分子(>10kb)移動速度接近， 很難分離形成足以區分的條帶。在脈沖場凝膠電泳中，電場不斷在兩種方向(有一定夾角，而不是相反的兩個方向)變動。DNA分子帶有負電荷，會朝正極移動。相對較小的分子在電場轉換后可以較快轉變移動方向，而較大的分子在凝膠中轉向較為困難。因此小分子向前移動的速度比大分子快。脈沖場凝膠電泳可以用來分離大小從10kb到10Mb的DNA分子。
CHEF-DR II脈沖場電泳系統

【介紹】
CHEF 基因組DNA 插塊試劑盒為PFGE 制備完整 的染色體大小的DNA 提供了方便。對于如此大的分子， 機械抽提會產生DNA 機械剪切, 導致了不好的PFGE 分離效果。要解決這個問題，通過將全細胞包埋于瓊 脂糖中進行裂解和去蛋白質，終獲得完整的純化 DNA。Bio-Rad 提供3 種試劑盒，分別用于制備細菌 （溶菌酶敏感）、哺乳動物的基因組DNA 及酵母染色體 （YACs）。每個試劑盒包括制備100 個插塊所需要的酶、 反應緩沖液和限制性酶切CleanCut 瓊脂糖，也包括一 次性插塊模和屏蔽帽以簡化插塊處理。每個試劑盒都 經過嚴格測試，保證所制備的基因組DNA 能被限制性 酶切并可在CHEF 電泳系統中分離。
