G1142 Weigert蘇木素染色液

產品說明： 
蘇木素是組織化學和免疫組織化學中常用的染料之一，可以廣泛用于組織切片或培養細胞的染色。本染色液可以和免疫熒光染色或免疫組化染色等配合使用。可以在蘇木素染色液染色后進行免疫熒光染色或其它 染料的染色，也可以在免疫組化染色后再進行蘇木素復染。染色步驟簡單，操作時間短。蘇木素染色后細胞核呈現藍色。

 使用說明: 
1、樣品處理 
a)對于石蠟切片：二甲***中脫蠟 5-10 分鐘。換用新鮮的二甲***，再脫蠟 5-10 分鐘。無水乙醇 5 分鐘。90%乙醇 2 分鐘。70%乙醇 2分鐘，蒸餾水 2 分鐘， 
b)對于冰凍切片：蒸餾水 2 分鐘， 
c)對于培養細胞：用 4%多聚甲醛固定 10 分鐘以上，蒸餾水洗滌 2 分鐘，換用新鮮的蒸餾水，再洗滌 2分鐘。  
2、蘇木素（HE）染色 
    對于上述處理好的樣品，用蘇木素染色 5-10 分鐘（可以根據染色結果和要求調整時間），用自來水洗去殘留的染色液，約 10 分鐘。蒸餾水再洗滌一遍(數秒鐘)。 
3、脫水、透明、封片或進行其它染色 
a)脫水、透明、封片： 95%乙醇脫水 2 分鐘，換用新鮮的 95%乙醇再脫水 2 分鐘；二甲***透明 5 分鐘，換用新鮮的二甲***，再透明 5 分鐘，用中性樹膠或其它封片劑封片。顯微鏡下觀察，細胞核呈藍色。 
b)進行其它染色： 
如果進行免疫熒光染色或熒光染料染色，在蘇木素染色后，70％乙醇洗滌 2 次，每次 2 分鐘。再用 PBS 或生理鹽水或 TBS或 TBST 等用于熒光染色的溶液浸泡 5 分鐘。 然后就可以進行免疫熒光染色或其它熒光染料的染色了。
